Correct your sequence to meet the String producibilty criteria

Use this function to make the sequence compliant with the String producibility criteria using the smallest possible set of silent mutations.

  1. Select your project in the Load tab.
  2. Click the Revise tab.
  3. Click the name of the sequence to display it in the central pane.
  4. In the Fix or Optimize pane, select the desired target organism from the Organism dropdown list.
  5. Select an ORF (open reading frame) or coding region from the ORF dropdown list or select Define Manually, then type the ORF.
  6. Click to apply the ORF selection.
  7. (Optional) Click Protect my cloning sites to define which sites to use for cloning, then select an appropriate cloning site present within the sequence, then click Save.

    Protect my cloning site is used to prevent:

    • A cloning site being removed during optimization

    • A cloning site sequence being changed during optimization

    • The algorithm from creating that cloning site anywhere else in the sequence during optimization

  8. (Optional) Click (Protected Regions) Add new to add regions that you do not want to be altered, then click Save.
  9. (Optional) Click (Motifs to avoid) Add new to add a motif that you do not want to be added to the sequence during optimization, then select the motif from the list, then click Save.
  10. In the Fix or Optimize pane, click Fix Problems.

    Note: Problem areas are highlighted and listed in the Problems pane and can also be corrected there by clicking Fix Sequence.
